ABSTRACT

<p><b>OBJECTIVE</b>To investigate the feasibility of using liquid chromatography (HPLC) to characterize the 3, 4-Dihydroxyphenylalanine (DOPA) redox state of mussel adhesive protein (MAP).</p><p><b>METHODS</b>The DOPA and protein contents of MAP were determined by HPLC, Arnow and Bradford methods respectively.</p><p><b>RESULTS</b>With extended oxidation time, the protein contents of MAP samples remained unchanged whereas the DOPA contents declined. The retention times of main peaks in HPLC for both the accelerated oxidation and retained samples shifted as the storage time extended, which could be related to the changes of sample redox state.</p><p><b>CONCLUSIONS</b>The redox state of MAP can be characterized by the change of HPLC peak retention time. HPLC can be used in the research on the MAP redox state, which is beneficial to the product development and quality control.</p>

ABSTRACT

Experiments on the preparation of Shenjincao (Palhinhaea cernua (L. )Franco et Vasc. jinjection by ultrafiltration was carried out. Innuence of pH and concentration on the quality of the prepared injection was investigated. It was found that the filterability of active principles was higher at lower pH and concentration,and vice versa. Therefore,a double ultrafiltration process was developed accordingly,which can remove macro and micro molecular impurities from the solution under different conditions. The quality of the injection was improved and loss of active principles was reduced to a minimum. Mechanism of the double ultrafiltration was briefly discussed.

ABSTRACT

Shenjincao injection was prepared from the extract of Lycopodium cernuum L. by ultrafiltration or wateralcohol methods,respectively,and then injected intra-peritoneally in groups of rats contaminated with quartz powder to assess their prophylactic effects. After 5 weeks of treatment,the rats were dissected and the fresh and dried weights,collagen content and the pathologic grading of the lungs were examined. Results showed that Shenjincao injection prepared with ultrafiltration was effective for the prophylactic treatment of experimental silicnsis of rats,while that prepared with water-alcohol method was devoid of such effect. The antisilicosis active principle of Shenjincao was explored and discussed.
